Abstract

Mucoadhesive patch is a pharmaceutical preparation which is designed to attach on the mucosal lining and able to prolong the drug contact time during application. Ethanolic extract of pomegranate rind (Punica granatum L.) shows inhibition activity to the bacterial and fungal growth which is commonly involved in oral and dental problems, thus, is expected to be used as an alternative (Stomatitis Aphthous Recurent (SAR)) medication. This study is focusing in the development of patch containing pomegranate rind extract with HPMC (Hydroxy Propyl Methyl Cellulose), PVP (Polyvinylpyrrolidone) and chitosan. The purpose of this research is to gain a mucoadhesive patch pomegranate rind extract with HPMC polymer, PVP and chitosan, as well as to determine its physical characteristics and stability. Extract of pomegranate rind was obtained by maceration method. Mucoadhesive patches containing pomegranate rind extract were prepared by solvent casting method. Formula was designed in six formula by varying the concentration of active substance the composition of HPMC, PVP and chitosan. Furthermore, the patches were physically evaluated including uniformity of weight, uniformity of dimensions, % swelling, weight loss at 30 minutes, weight loss at 60 minutes, pH of the patch surface, adhesion time and folding endurance. Data was were analyzed using SPSS . The results showed that the ethanolic extract of pomegranate rind was able to be formulated into mucoadhesive patches with polymer HPMC, PVP and chitosan. The results of the physical charateristic test showed that F6 has the best characteristics compared to F1, F2, F3, F4, and F5 with uniformity of weight (21 ± 0.94 mg), uniformity of dimensions (0.44 ± 0.03 mm), value of % swelling (97.91 ± 23.80%), weight loss at 30 minutes (0.05 ± 0.09), weight loss at 60 minutes (0.32 ± 0.09), surface pH patch (3.25 ± 0.09 ), mucoadhesion time (59.57 ± 54.45 min) and folding endurance test (2 folds). All the results of characteristic test is ideal, unless the surface pH and folding endurance test. The results of the physical stability test and SPSS analysis showed patch changes during storage within 7 days of probability values P <0.05.
