Abstract

Cervical cancer is a malignant neoplasm of the cervical region, is the third leading cause of cancer deaths among women. Waru (Hibiscus tiliaceus L.) leaves contain flavonoid compounds and showed cytotoxic effect against gastric cancer cells, colon cancer cells, and breast cancer cells which has potentially as a chemopreventive agent. This study is aimed at determining the antioxidant and cytotoxic activity of FEDW in HeLa Cervical Cancer Cell Line and in silico tested using molecular docking. Waru leaves powder extraction was done by maseration using ethanol 70%, then fractionated liquid-liquid extraction with ethanol and chloroform. The study of contain from the ethanol fraction of waru leaves was done by thin-layer chromatography (TLC) method. Antioxidant activities of FEDW was done by DPPH method compared with rutin as a positive control. Cytotoxicity activities of FEDW was done by MTT Assay. To determine the interaction of rutin against protein targets Bcl-xl tested in silico with PLANTS (Protein Ligand ANT System) program. The result of thin-layer chromatography (TLC) showed that fraction ethanol of Waru leaves consist of flavonoid, The results used DPPH method and rutin as positive control showed that FEDW have antioxidant activities with value of 28 μg/ml and had been potential to be cytotoxic againts HeLa cells with IC50 value of 1605 μg/ml. The result on molecular docking showed that rutin compound have strong potential in inhibited protein Bcl-xl in molecular docking with a docking score of -91.210. In conclusion FEDW have antioxidant activity and have anticancer activity were classified as less toxic, and has inhibitory interactions of protein Bcl-xl.
