Abstract
Phenolic is a class of secondary metabolite compounds contained in moringa (Moringa
oleifera L.) and winged bean (Psophocarpus tetragonolobus L.) plants. The phenolic content
contained in moringa seeds and winged bean seeds has antibacterial properties. The purpose of
this study was to compare total phenolic content and antibacterial activity of the ethyl acetate
fraction from Moringa seed (Moringa oleifera L.) and winged bean seed (Psophocarpus
tetragonolobus L.) against Staphylococcus aureus and Escherichia coli bacteria.
Moringa seed and winged bean seed powder was extracted by maceration method using
96% ethanol. The viscous extract obtained was fractionated using the liquid-liquid method in
stages using n-hexane, ethyl acetate and ethanol: water as solvents. The fractions obtained were
identified for phytochemical content using a color test and the results showed that the moringa
seeds and winged bean seeds were positive for phenolic, alkaloid and saponin content. Total
phenolic testing was carried out using the Folin-ciocalteau method on the ethyl acetate fraction
of Moringa seeds and winged bean seeds. Antibacterial activity testing was carried out using
direct testing and literature review studies.
The results showed that the total phenolic content of winged bean seeds was greater than
that of moringa seed with levels obtained of 43.16 ± 0.404 mgGAE / g for moringa seed and
230.1 ± 12.586 mgGAE / g for winged bean seed. Based on the direct test results, the ethyl
acetate fraction of the ethanol extract of winged bean seed has a strong inhibitory power
category at a concentration of 22.5% against Escherichia coli bacteria. Based on the results of
the literature review, winged bean seed have antibacterial activity which is more effective
against Staphylococcus aureus bacteria. Meanwhile, Moringa seed are more effective in
inhibiting Escherichia coli bacteria.
