Abstract

Paracetamol is one of drug which can cause hepatic necrosis directly. Until now, society doesn't realize with this condition so the epidemiology become higher. Back to nature treatment using Nigella sativa is one of choice to cure hepatic necrosis. This research aims to determine the curative effect of N.sativa ethanolic extract to histological structure of paracetamol-induced hepatic necrosis on white rat.
This research use laboratorium experimental with post-only control group design. Research subject use 2-3 months old male white rat (Rattus norvegicus) Sprague Dawley which is divided into negative control, paracetamol control, 200 mg/kgBW of extract, 400 mg/kgBW of extract, and 800 mg/kgBW of extract. 1000 mg/kgBW of paracetamol is given during 7 days followed by extract giving during 9 days. Histological structure of liver is observed to know liver destruction and inflammation degree. Statistical analyzing test uses Kruskal-Wallis continued with Mann-Whitney test. -The result of statistical analyzing shows that there is difference between group (p<0,001) and extract group is proven can make histological structure better (p<0, 001). Between 200 mg/kgBW and 800 mg/kgBW {p=O, 014 (inflammation degree) and p-0,004 (liver destruction degree), there is significantly different. But, -there is no significantly difference between 200 mg/kgBW and 400 mg/kgBW of extract dose fp-0,838 (inflammation degree) and 0,306 (liver destruction degree)] and between 400 mg/kgBW and 800 mg/kgBW of extract dose {p-0, 064 (inflammation degree) and 0,054 (liver destruction degree)]. The conclusion is that giving 200 mg/kgBW, 400 mg/kgBW, and 800 mg/kgBW of N.sativa ethanolic extract during 9 days has a curative effect to paracetamol-induced hepatic necrosis which is indicated by a better inflammation and liver destruction degree.
