Abstract
Paracetamol is one of drug which can cause hepatic necrosis directly. There is higher amount of liver necrosis caused by hepatotoxic drugs consumption. Until now, society doesn't realize with this condition so the epidemiology become higher. Back to nature treatment using Nigella sativa is one of choice to prevent hepatic necrosis. This research aims to determine the preventif effect of Nigella sativa ethanolic extract to paracetamol-induced hepatic necrosis on white rat by measuring SGPT level. Research design use laboratory experiment with post only control group design. The subject are 2-3 months male white rats (Rattus norvegicus), Sprague Dawley, divided into negative control group, 1000 mg/kgBW paracetamol control group, 200 mg/kgBW extract dose treatment group, 400 mg/kgBW extract dose treatment group, and 800 mg/kgBW extract dose treatment group. Etanolic extract of N sativa in appropriate dosage group given for 16 days, followed by paracetamol 1000 mg/kgBW start from 10th day for 7 days. SGPT score measured with spectophotometer then analized with Parametric Test One Way Anova. SGPT rate in control negative group was measured 44,95±2,62 IU/L; in paracetamol control group was 68,3015,09 IU/L; in 200 mg/kgBW extract dose treatment group was 68,3015,09 IU/L; in 400 mg/kgBW dose was 55,40±8,19 IU/L; in 800 mg/kgBW dose was 40,45±12,08 IU/L. Statistical test show p = 0,007 or p < 0,05 means that there is significance different one group another. It can be concluded that 16 days N. sativa seed ethanolic extract administration (200 mg/kgBW, 400 mg/kgBW, 800 mg/kgBff9 followed by paracetamol 1000 mg/kgBW start from 10th day for 7 days on Rattus norvegicus decrease SGPT score.
