Abstract

Background: Relapse is a post-orthodontic treatment problem caused by the physiologic response of tooth-supporting tissues resulting the teeth to move back to the pre-treatment position. Pharmacological retention with anti-resorptive drugs such as bisphosphonate risedronate can be used to prevent relapse. Bisphosphonate can stimulate osteoblast proliferation, which is cells that form bones. Bone formation after orthodontic treatment is an essential part of tooth retention thus preventing relapse. Emulgel form is chosen as the drug carrier because of its easy topical application and local effect. Objective: This study aims to determine the effect of bisphosphonate risedronate in emulgel form on osteoblast count. Methode: A total of 27 male guinea pigs ± 8-12 week-old, ±500-600 grams are divided into three groups, which are control group or group without drug application (I), emulgel without risedronate (II), and emulgel with risedronate (III). Open coil springs were applicated on the guinea pig inter incisor for 14 days with 7 days of stabilization. During the stabilization phase, emulgel without risedronate and emulgel with risedronate were begun to be given to group II and III. The orthodontic appliance was removed and the application of emulgels continued. Samples were decapitated and histologic spesicem were made with HematoxylinEosin to measure osteoblast count at day 0, 3, and 7 after the orthodontic appliance was removed. One way ANOVA test was used with SPSS 23 to determine the differences among groups. Result: The emulgel with risedronate group showed the highest osteoblast count on day 3 and 7 (p<0,05). LSD test showed a significant difference between emulgel with risedronate groups with control and emulgel without risedronate group (p<0,05). Conclusion: Application of bisphosphonate risedonate is effective to increase osteoblast count on day 3 and 7.
