Abstract

Colon cancer is cancer that attacks the colon with a relatively high mortality rate in the world, including in Indonesia. The problems that arise from cancer treatment using chemotherapy are not selective and can cause severe side effects. Melinjo (Gnetum gnemon L.) seeds are reported to contain stilbenoid compounds such as gnetin C, resveratrol, and isorhapontigenin which have anticancer activity. This study aims to determine the activity of the Melinjo Seeds Ethyl Acetate Fraction against colon cancer cells WiDr by in vitro and in silico study. Melinjo seeds powder was extracted by maceration method using 70% ethanol then fractionated using ethyl acetate as a solvent to obtain Melinjo Seeds Ethyl Acetate Fraction (MSEAF). The compounds in MSEAF was identified using the LC-MS method. In silico test using the bioinformatics method to determine the target proteins of the compounds and molecular docking test using Autodock Vina to determine the binding affinity between the compounds with target proteins. In vitro cytotoxic test on colon cancer cells WiDr were carried out using the MTT assay method, then continued with the cell cycle test using flowcytometry method. The results showed that MSEAF contained gnetin C, gnemonoside D, and gnemonol M which is a stilbenoid compounds. Target proteins that obtained from bioinformatics test of gnetin C compound was MAPK3 and KRAS. Gnetin C and MAPK3 protein had the best binding affinity in the molecular docking test with a docking score of -11,2 kcal/mol.
