Abstract
Background: Lung cancer is the leading cause of cancer death worldwide. Various anticancer therapies have been developed, one of which is targeted therapy. However, currently resistance to targeted therapy has emerged. Lactococcus lactis is one of the lactic acid bacteria that has the potential as an anticancer. The purpose of this study was to determine the antiproliferative activity of Lactococcus lactis bacterial extract against HTB-179 lung cancer cells. Methods: Cytotoxicity test was carried out by giving several series of concentrations of the supernatant and pellets of Lactococcus lactis crude extract of 1000; 500; 250; 125; and 62.5 g/ml. The test was carried out using the MTT Assay method, then the percent death was calculated and the IC50 value was obtained. The study was continued with an antiproliferative test to determine the doubling time of the samples treated with IC50, IC50 levels and control cells at 0, 24, 48, and 72 hours. Results: The results showed that the supernatant of Lactococcus lactis crude extract had a cytotoxic effect on HTB-179 lung cancer cells with an IC50 value of 199.27 g/ml, while the pellets of Lactococcus lactis crude extract had no cytotoxic effect on HTB-179 lung cancer cells, so it cannot be continued for antiproliferative test. The results of the antiproliferation test showed that there was an inhibition of the growth of HTB-179 lung cancer cells in the supernatant of Lactococcus lactis crude extract treatment with a doubling time value of 16 hours in control cells, 23 hours in the treatment with a concentration of IC50 (99.635 g/mL), and 27 hours in the treatment with a concentration of IC50 (199.27 g/mL). Conclusion: Supernatant of Lactococcus lactis crude extract in the form of had antiproliferative effect, while pellets of Lactococcus lactis crude extract did not have antiproliferative effect on HTB-179 lung cancer cells.
