Abstract
Acne is a common skin problem that bothers and often occurs, caused by bacteria called Staphylococcus aureus. Antibiotics such as tetracycline, erythromycin and clindamycin are often used for acne treatment, however, this can cause skin irritation and develop resistance to antibiotcis, so treatment with natural ingredients is chosen. The purpose of this study was to develop natural ingredients as an alternative to acne treatment due to bacterial infection from a combination of propolis extract and Moringa leaf extract in the form of a facial serum, and a phytochemical screening test was carried out on the extract to see the secondary metabolite content as antibacterial. The method used in this research is an experimental laboratory. Propolis and Moringa leaf extracts were tested for flavonoid identification to determine the presence of flavonoid content using the Willstatter method followed by a TLC test with the results of the flavonoid identification test proven to contain flavonoid in both extracts with a color change to yellow after the addition of concentrated Mg and HCL powder. However, the TLC test with comparison of quercetin did not show any similarity in the stains on the two extracts, which indicated that the propolis and Moringa leaf extracts in this study did not contain quercetin-type flavonoid compounds. Furthermore, the formulation of propolis extract and Moringa leaf extract were made in the form of facial serum preparations, then antibacterial testing was carried out against Staphylococcus aureus. Antibacterial test was carried out using the disc diffusion method. The results obtained in F1 are serum with a concentration of 10% Propolis and 4% Moringa, the average diameter of the inhibition zone formed is 2.33±0.25 mm, at F2 is 30% Propolis and 5% Moringa is 2.7± 0.44 mm and at F3 that is 50% Propolis and 6% Moringa 2.67±0.35 mm. Positive control, namely tetracycline antibiotics, the average diameter of the inhibition zone formed was 17±0 mm, while in negative control, serum without extract was 0±0 mm. Further analysis was performed using the Kruskal-Wallis test followed by the Mann-Whitney test. Based on the Mann-Whitney test on the three concentration variants, there was no significant difference in inhibiting the growth of Staphylococcus aureus bacteria. The conclusion of this study is that there is antibacterial activity shown in the formulation of facial serum preparations with a combination of propolis extract and Moringa leaf extract in inhibiting the growth of acne-causing bacteria, namely Staphylococcus aureus, which is classified as weak compared to positive control, namely tetracycline antibiotics. The presence of antibacterial activity in propolis and Moringa leaf extracts is due to the content of secondary metabolites, namely flavonoid compounds as antibacterial.
