Abstract
The skin is one of the largest organs in the human body and consists of tissues that have a specific function as a protector and cover that guards other organs of the body. Human skin can undergo physiological changes with age, resulting in aging of the skin. There are several types of factors that cause aging on the skin such as intrinsic and extrinsic factors. In previous studies, it was known that parijoto fruit has antioxidant activity. Parijoto fruit contains polyphenol compounds that can improve the balance of ROS (Reactive Oxygen Species) with mechanisms such as inhibiting free radicals. This study aims to find out the biological activity of Medinilla speciosa fruit, Reinw.ex Bl as an active anti-aging ingredient in vitro. The process of making the extract begins with the softening method using 70% ethanol solvent. In addition, phytochemical screening and identification of secondary metabolites by the KLT method. As a comparison, a test of antioxidant activity was conducted with the DPPH method using quercetin. This is continued by the activity test in vitro anti-aging activity test, i.e. a cell viability test using the adult MTT Assay Human Dermal Fibroblast (HDFa) test, and an elastase enzyme inhibition test using Human Neutrophil Elastase (HNE). The results showed that EEBP produced a viscous extract weighing 94.23 grams. EEBP contains flavonoid polyphenol compounds through the KLT method and contains secondary metabolites in the form of saponins, tannins, and flavonoids on phytochemical examination. The antioxidant activity of EEBP with an IC50 value obtained of 122.19 μg / mL is categorized as a powerful antioxidant and quercetin as a comparison with an IC50 value of 6.25 μg / mL is categorized as a very strong antioxidation. EEBP can inhibit the activity of the enzyme elastase. HDFa cell viability test results show that EEBP can affect HDFa cell viability. From the results of this study, it can be concluded that parijoto ethanol extract (Medinilla speciosa, Reinw. ex Bl) has the potential to be an active ingredient in antiaging cosmetics.
