Abstract
In previous studies, hibiscus leaves contain flavonoids, saponins and polyphenols. One of the compounds contained in the leaves can be used as an antioxidant. Antioxidants are included in compounds that can prevent the spread of free radicals. This study aims to determine the compound content in the ethyl acetate fraction of ethanol extract and the antioxidant activity of hibiscus leaves based on the DPPH (1,1-Diphenyl-2-Picrylhydrazyl) test which is expressed in IC50. Fractionation is done to separate compounds based on their level of polarity. Ethyl acetate can attract polar and non-polar compounds because it is semi-polar so that when fractionation is carried out, more hibiscus leaf compounds are attracted. This research is an experimental quantitative and qualitative research laboratory analysis of compounds that have antioxidant activity in the ethyl acetate fraction of hibiscus leaves. The method used is phytochemical test and antioxidant activity test using DPPH by calculating the IC50 value. The reaction between DPPH and the sample will remove the purple color of DPPH as an indicator of antioxidant activity The results of the study seen from IC50 which presented antioxidant activity showed that the ethyl acetate fraction of hibiscus leaf ethanol extract had a value of 119.26 ppm and had moderate criteria. The ethyl acetate fraction of hibiscus leaf ethanol extract was positive for chemical compounds of flavonoids and tannins, which was proven by phytochemical testing that there was a color change in the sample.
