Abstract
Phalaenopsis amabilis is a native plant of Indonesia that is found in great diversity in every Indonesian forest so Phalaenopsis amabilis is designated as the “National Flower of Indonesia”. Based on Presidential Decree No. 4 of 1993 Phalaenopsis amabilis as “Puspa Pesona Indonesia”. The diversity of Phalaenopsis amabilis in Indonesia is not widely known, so moleculer reseach is needed with the first step, namely DNA isolation. DNA isolation can be carried out using the CTAB method and using a kit. The purpose of this study was to obtain the quantity and quality of DNA isolation result from the Phalaenopsis amabilis based on a comparison of the use of two different methods, namely CTAB dan kit. The reseach method used was a single factor experiment with a combination of sample weight and incubation time consisting of 9 treatments with 2 replications, so that 18 experimental units were obtained. Phalaenopsis amabilis with DNA isolation using the CTAB method which had a sample weight of 0,25 grams, 0,5 grams, and 0,75 grams, and each sample weight was incubated for 30 minutes and 60 minutes. In addition, there is a DNA isolation treatment using a kit that has a sample weight of 0,25 grams, 0,5 grams, and 0,75 grams and incubation for 10 minutes. The results showed that the isolation of Phalaenopsis amabilis crown DNA using the CTAB method produced a good DNA quality compared to using the kit and the best treatment was at 0,75 g for 30 minutes with a consentration of 850,445 ng/µl and purity of 2,0.
