Abstract
Parijoto (Medinilla speciosa (Reinw ex BL) Bl) is a plant that is widely used traditionally by the community as anti-cholesterol, anti-bacterial, and anti-diarrhea. Based on previous research, parijoto fruit contains secondary metabolites of flavonoids, alkaloids, tannins, saponins, glycosides and terpenoids. This study aims to determine the potential immunomodulatory activity in vitro possessed by parijoto fruit and aims to determine the flavonoid content contained in parijoto fruit using the Thin Layer Chromatography method and determine the total flavonoid levels contained therein. The test was carried out by extraction using the maceration method with 70% ethanol solvent. The thick ethanol extract obtained was then tested on a phytochemical screening for the secondary metabolites of flavonoids using the TLC method using routine and quercetin as comparisons. Furthermore, the total flavonoid levels were determined using the colorimetric method. The immunomodulatory activity test was seen from the phagocytosis activity and lymphocyte proliferation. The phagocytic activity of macrophages was measured using the phagocytosis index and phagocytic capacity parameters. Lymphocyte proliferation activity using the MTT Assay method and measured using the proliferation stimulation index parameter. The results of phytochemical screening using the TLC method showed that the ethanol extract of parijoto fruit contained secondary metabolites of flavonoids. The ethanol extract of parijoto fruit had a total flavonoid content of 7.74 ± 0.17 QE /g sample. The immunomodulator test results in vitro showed that the ethanol extract of parijoto fruit could increase the phagocytosis activity of macrophages compared to control cells. A concentration of 500 μg/mL showed phagocytosis activity of macrophages which had significant differences compared to other concentrations of control cells with a Phagocytosis Index value of 4.65 and a value of 76% Phagocytic Capacity. Lymphocyte proliferation test obtained an IS value of 3 which showed a positive effect on proliferation. This study shows that the ethanol extract of parijoto fruit can increase the activity of macrophage phagocytosis and lymphocyte proliferation.
