Abstract

Orchid Phalaenopsis amabilis is one of the 3 national flowers designated by the government as an Indonesian charm flower. Moon orchid cultivation conventionally with the separation of keiki has a long propagation time, thereforeculture propagation is carried out in vitro. This study aims to determine the optimum concentration of BAP and NAA on shoot growth in the leaf culture of the moon orchid in vitro. This study was conducted using an experimental method arranged in a single-factor completely randomized design consisting of 6 treatments, namely NDM 1 mg/l BAP + 0 mg/l NAA, NDM + 1 mg/l BAP + 0.5 mg/l NAA, NDM + 2 mg/l BAP + 0 mg/l NAA, NDM + 2 mg/l BAP + 0.5 mg/l NAA, NDM + 3 mg/l BAP + 0 mg/l NAA, and NDM + 3 mg/l BAP + 0.5 mg/l NAA. Each treatment was repeated 9 times. The results of this study showed that the administration of BAP and NAA with a concentration of 2 mg/l BAP + 0.5 mg/l NAA on NDM medium was the best treatment in inducing shoots in orchid leaf explants of the moon orchid. with the percentage of explants sprouting at 22.22% and the number of explants sprouting as much as 2. These results need further research in the administration of BAP and NAA on a medium other than NDM in an effort to induce shoots in order to determine the effect of shoot growth other than BAP and NAA.
