Abstract
Vanda tricolor is an orchid endemic to Mount Merapi. V. tricolor var. Suavis which is located on the slopes of Mount Merapi will be threatened by the eruption of Mount Merapi. Several attempts have been made to preserve the Vanda tricolor orchid, one of which is through in vitro culture. In vitro culture studies of Vanda tricolor have been carried out and large numbers of shoots have been obtained through multiplication and then raised by subculture. Acclimatization is the adaptation period of bred plants in in-vitro culture whose conditions were initially controlled and then changed to field conditions whose conditions were no longer under control. Growing medium and fertilizer application are important factors in the acclimatization process. This study aims to determine the best type of medium and fertilizer for acclimatization of Vanda tricolor orchids. This research was conducted using an experimental method in the preparation of a Factorial Completely Randomized Block Design (RAKL) consisting of the first factor being the type of medium, namely coconut coir, moss and coconut coir + moss and the second factor was the type of fertilizer, namely no fertilizer, leaf fertilizer and POC. From the two treatment factors, 9 treatment combinations were obtained. Each treatment was repeated 3 times and each replication consisted of 3 samples. The results showed that the administration of coconut coir + moss medium was the best treatment for increasing plant height and leaf length. The application of Gandasil D Leaf fertilizer is the best treatment for increasing plant height. While offering POC fertilizer is the best treatment for leaf length.
