Abstract
This study aimed to determine effect and best concentration of combination from
2,4-Diclorofenoxyacetic acid and cytokines in medium of liquid organic fertilizer on
somatic embryogensis Vanda tricolor. This research was held on January – March 2020.
This study used the experimental methods, arranged in a Completely Randomized
Design. There are 6 treatments, with 3 replication and on each replication there are 3
samples, including 2 level concentration of 2,4-D (1 mg/l and 2 mg/l) and sources of
cytokines (Thidiazuron 0,1 mg/l and coconut water 150 mg/l) The parameters include
percentage of viable explant, percentage of browning explant, percentage of
contamination explant, percentage of vitrification explant, shoot height increase, number
of leaves increase, leaf colour, time to form proembryo, number of proembryo, and
developmental morphology of the shoot. Analyzed by variance analysis (ANOVA) and
DMRT test at 5% level.
For analysis result of time to form proembryo and number of proembryo showed
that the interaction on each treatment gave no significant difference. However, the result
of analysis showed that there are effects between Liquid Organic Fertilizer (POC) with
2,4-D and coconut water 150 ml/l. Treatment medium using POC+2,4-D 1 mg/l+coconut
water 150 ml/l proved to gave appearance of proembryo and induces the increase of the
number of proembryo (1,89 proembryo).
