Abstract
Colorectal cancer ranks third in the world as the most suffered cancer. Based
on previous research, flavonoid compounds in leaves of dandang gendis
(Clinacanthus nutans L.) are useful as anticancer and antioxidants. Previous
research shows that herbal dandang gendis has inhibitory activity on breast cancer
cells HeLa. This study aims to determine the potential content of flavonoids,
namely luteolin, one of which is in the form of isoorientin in the ethanol fraction of
herbal dandang gendis (Clinacanthus nutans L.) (FEHDG) as a co-agent
chemopreventive against WiDr colorectal cancer cells, and compared with
conventional drugs doxorubicin.
In this research begins with extraction by maceration using 70% ethanol.
Fractionation by the liquid-liquid method and the solvent solution used is ethyl
acetate. In vitro methods for antioxidant testing are the DPPH test (2,2-diphenyl-1-
picrylhydrazyl) and the cytotoxic test using the MTT Assay method on WiDr colon
cancer cells, followed by co-chemotherapy with doxorubicin. In in silico test used
molecular docking on isoorientin compounds with IKK and VEGF as the target
protein and compared with doxorubicin.
Antioxidant test results showed a weak ability with IC50 value of FEHDG is
2556.62 μg/mL. In the cytotoxic test FEHDG had less active cytotoxic activity with
an IC50 value is 1737.9 μg/mL. FEHDG has a strong effect of co-chemotherapy,
with CI value is 0.11 at the best combination, on a concentration of FEHDG 108.56
µg / mL (1
/16 IC50) and Doxorubicin 0.108 (1
/16 IC50) µg/mL. Isoorientin compounds
have strong bonds in the IKK and VEGF proteins with docking score -9.7 kcal/mol
and -6.9 kcal/mol.
