Abstract
Breast cancer is ranked the second highest cause of death in Indonesia.
Cancer treatment has the goal to extend expectations and improve the quality of life
of sufferers. Of the several cancer treatment options available, it often causes severe
side effects for patients. Tin (Ficus carica L.) leaves contain ingredients that can
function as anticancer. The purpose of this study was to identify secondary
metabolite compounds present in Tin leaves, test the antioxidant and cytotoxic
activity of MCF-7 cells from ethanol extract, n-heksan fraction and ethyl acetate
fraction from Tin leaves.
Tin leaf powder macerated with 70% alcohol for 7 days. The extracts were
then fractionated using n-heksan, and ethyl acetate. Identification test with ethanol
extract was then carried out using spray reagents. Other identification tests were
also conducted using the literature study method. Ethanol extract, fraction n-heksan
and ethyl acetate were then tested for antioxidant activity by DPPH method and
cytotoxic test using MTT method.
The results of identification with TLC and literature studies found that the
ethanol extract of Tin leaves contains secondary metabolite compounds in the form
of flavonoids, alkaloids and steroids. Based on the results of literature studies, Tin
leaves have antioxidant activity ranging from very strong to weak depending on the
type of variety and treatment.. The ethyl acetate fraction of Tin leaf is thought to
have weak cytotoxic properties with an IC50 value of 274.5877 μg / mL while the
ethanol extract and n-heksan fraction of the Tin leaf do not have cytotoxic
properties because it has an IC50 value of 562,827; 576,3552 μg / mL. Based on
the research results Tin leaves have the potential to be developed as a
chemopreventive agent for MCF-7 breast cancer cells.
